Infection-Responsive Expansion of the Hematopoietic Stem and Progenitor Cell Compartment in Zebrafish Is Dependent upon Inducible Nitric Oxide
Using a zebrafish model of demand-driven granulopoiesis to explore the hematopoietic stem and progenitor cell (HSPC) response to infection, investigators present data supporting a model where the zebrafish ortholog of the cytokine-inducible form of nitric oxide synthase (iNOS/NOS2) Nos2a acts downstream of the transcription factor C/ebpβ to control expansion of HSPCs following infection. The Combination of Valproic Acid and Lithium Delays Hematopoietic Stem/Progenitor Cell Differentiation
The ability to inhibit hematopoietic stem/progenitor cell commitment in culture may be of benefit to cell therapy protocols. Here, investigators tested two small molecules, valproic acid and lithium to inhibit differentiation. [Blood] Abstract
Differential Role of Actin-Binding Proteins in Controlling the Adipogenic Differentiation of Human CD105-Positive Wharton's Jelly Cells
Wharton's Jelly cells subjected to adipocyte induction were observed for changes in the cell morphology and alterations in actin filament formation. [Biochim Biophys Acta] Abstract
Characteristics of Thawed Autologous Umbilical Cord Blood
Using a standardized thaw-wash method, autologous umbilical cord blood samples were assessed for viable total nucleated cells (vTNCs), viable CD34+, and colony-forming unit-granulocyte-macrophage counts. Postthaw %vTNC recoveries were compared against processing characteristics and analyzed according to processing method, cryopreservation volume, concentration, container, and length of storage. [Transfusion] Abstract
Cryopreservation of Umbilical Cord Blood with a Novel Freezing Solution that Mimics Intracellular Ionic Composition
This study evaluated the preservation efficacy of a novel intracellular-like cryopreservation solution, the rate of addition of two cryopreservation solutions to cord blood units, and reduced final dimethyl sulfoxide concentration of 5%. [Transfusion] Abstract
Early CD3 Peripheral Blood Chimerism Predicts the Long-Term Engrafting Unit following Myeloablative Double Cord Blood Transplantation
Beginning at day seven post-transplant, researchers prospectively measured weekly lineage-specific peripheral blood donor chimerisms in patients undergoing myeloablative double cord blood transplantation to evaluate whether the degree of early donor contribution to specific lineage(s) would predict the long-term engrafting unit. [Biol Blood Marrow Transplant] Abstract
Myeloablative Transplantation Using Either Cord Blood or Bone Marrow Leads to Immune Recovery, High Long-Term Donor Chimerism and Excellent Survival in Chronic Granulomatous Disease
Twelve consecutive children with severe chronic granulomatous disease received myeloablative hematopoietic stem cell transplantation using sibling bone marrow, unrelated cord blood, and sibling cord blood. [Biol Blood Marrow Transplant] Abstract
Human Cord Blood-Derived Multipotent Stem Cells (CB-SCs) Treated with All-Trans-Retinoic Acid (ATRA) Give Rise to Dopamine Neurons
To explore their therapeutic potential in Parkinson’s disease, researchers examined whether CB-SCs could be induced to differentiate into dopamine neurons in the presence of ATRA. [Biochem Biophys Res Commun] Abstract
