LABORATORY RESEARCH
The Common Feature of Leukemia-Associated IDH1 and IDH2 Mutations Is a Neomorphic Enzyme Activity Converting Alpha-Ketoglutarate to 2-Hydroxyglutarate Researchers report that tumor 2-hydroxyglutarate is elevated in a high percentage of patients with cytogenetically normal acute myeloid leukemia. [Cancer Cell]
Induction of Cell Cycle Entry Eliminates Human Leukemia Stem Cells in a Mouse Model of Acute Myeloid Leukemia Using a NOD/SCID/IL2r(gamma)null mouse model of human acute myeloid leukemia (AML), researchers show that the AML stem cells in the endosteal region are cell cycle quiescent and that these stem cells can be induced to enter the cell cycle by treatment with granulocyte colony-stimulating factor. [Nature Biotechnol]
Apoptosis Induced by JAK2 Inhibition is Mediated by Bim and Enhanced by the BH3 Mimetic ABT-737 in JAK2 Mutant Human Erythroid Cells JAK2 inhibition-induced apoptosis correlated with up-regulation of the non-phosphorylated form of the BH3-only protein Bim in hematopoietic cell lines bearing JAK2 mutations. [Blood]
Deletion of Puma Protects Hematopoietic Stem Cells and Confers Long-Term Survival in Response to High-Dose Gamma-Irradiation Researchers have examined the role of p53 up-regulated mediator of apoptosis (PUMA) in apoptosis of hematopoietic stem cells after radiation injury. [Blood]
Integrated Analysis of MicroRNA and mRNA Expression Profiles in Physiological Myelopoiesis: Role of hsa-mir-299-5p in CD34+ Progenitor Cells Commitment Gain- and loss-of-function experiments revealed that hsa-miR-299-5p participates in the regulation of hematopoietic progenitor fate, modulating megakaryocytic-granulocytic versus erythroid-monocytic differentiation. [Cell Death Differ]
Standards, Regulations and Accreditation for Registries Involved in the Worldwide Exchange of Hematopoietic Stem Cell Donors and Products The World Marrow Donor Association is working closely with other international organizations working in cellular therapy such as the Worldwide Network for Blood and Marrow Transplantation to develop and maintain global recommendations and requirements for a standardized practice. [

Increased Levels of NOTCH1, NF-KappaB, and Other Interconnected Transcription Factors Characterize Primitive Sets of Hematopoietic Stem Cells
To further evaluate the molecular basis related to the more primitive characteristics of umbilical cord blood (UCB) and CD133+ hematopoietic stem cells, immunomagnetically purified human CD34+ and CD133+ cells from bone marrow and UCB were used on gene expression microarrays studies. CLINICAL RESEARCH
Phase I Study of KW-0761, a Defucosylated Humanized Anti-CCR4 Antibody, in Relapsed Patients With Adult T-Cell Leukemia-Lymphoma and Peripheral T-Cell Lymphoma KW-0761, a defucosylated humanized anti-CC chemokine receptor 4 (CCR4) antibody, exerts a strong antibody-dependent cellular cytotoxic effect. This phase I study assessed the safety, pharmacokinetics, recommended phase II dose and efficacy of KW-0761 in patients with relapsed CCR4-positive adult T cell leukemia-lymphoma or peripheral T cell lymphoma. [J Clin Oncol]
Favorable Outcome in Patients with Acute Myeloid Leukemia with NPM1 Mutation Autografted After Conditioning with High Dose Continuous Infusion Idarubicin and Busulphan
This study suggests that BuI regimen (based on high dose continuous infusion idarubicin and busulphan) results in favorable clinical outcome in patients with isolated NPM1 mutation and could be investigated in a randomized study versus other regimes or repeated courses of high dose cytosine-arabinoside. [Biol Blood Marrow Transplant]
Prognostic Significance of Genetic Variants in the IL-23/Th17 Pathway for the Outcome of T Cell-Depleted Allogeneic Stem Cell Transplantation Results suggest the potential usefulness of IL-23/Th17 genotyping in donor selection and patient evaluation. [Bone Marrow Transplant]
