Blood Stem Cell, Leukemia Link Illuminated in UCSF-Led Study
A UCSF-led team has discovered at least one key reason why blood stem cells are susceptible to developing the genetic mutations that can lead to adult leukemia. [Press release from the University of California discussing online prepublication in Cell Stem Cell]
Cancer's 'Addiction' Spurs New Treatment Hopes
Cancer can call upon a bewildering array of genetic tricks to wreak havoc, but researchers show that the disease can become dependent upon a tiny gene that allows it to adapt and proliferate. [Press release from Yale Cancer Center discussing online prepublication in Nature]
Chronic Health Conditions Common for Stem Cell Transplant Survivors
Although hematopoietic cell transplantation cures many blood diseases, two-thirds of long-term survivors report at least one chronic health condition after the procedure, according to a recent study. [Press release from the American Society of Hematology discussing online prepublication in Blood]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal)
LABORATORY RESEARCH
OncomiR Addiction in an In Vivo Model of MicroRNA-21-Induced Pre-B-Cell Lymphoma
Results demonstrate that tumors can become addicted to oncomiRs and support efforts to treat human cancers through pharmacological inactivation of microRNAs such as miR-21. [Nature]
Therapeutic Cell Engineering with Surface-Conjugated Synthetic Nanoparticles
Here, researchers describe a strategy to enhance cell therapy via the conjugation of adjuvant drug-loaded nanoparticles to the surfaces of therapeutic cells. [Nat Med]
Inducible fli-1 Gene Deletion in Adult Mice Modifies Several Myeloid Lineage Commitment Decisions and Accelerates Proliferation Arrest and Terminal Erythrocytic Differentiation
This study investigated the role of the ETS transcription factor Fli-1 in adult myelopoiesis using new transgenic mice allowing inducible fli-1 gene deletion. fli-1 deletion in adult induced mild thrombocytopenia associated with a drastic decrease in large mature megakaryocytes number. [Blood]
BMI1 Collaborates with BCR-ABL in Leukemic Transformation of Human CD34+ Cells
Here, researchers show that BMI1 collaborates with BCR-ABL in inducing a fatal leukemia in NOD-SCID mice transplanted with transduced human CD34+ cells within 4-5 months. [Blood]
Dose-Dependent Regulation of Primitive Erythroid Maturation and Identity by the Transcription Factor Eklf
Researchers conclude that Eklf gene dosage is crucial for regulating the surface phenotype and molecular identity of maturing primitive erythroid cells. [ Blood]
Developmentally Induced Mll1 Loss Reveals Defects in Postnatal Hematopoiesis
Researchers show that VavCre;Mll mutants exhibit phenotypically normal fetal haematopoiesis, but rarely survive past three weeks of age. [Leukemia]
Involvement of the SRF Coactivator Megakaryoblastic Leukemia in the Activin-Regulated Dendritic Complexity of Rat Cortical Neurons
Here, researchers individually knocked down the four transcription factors PU.1, IRF8, MYB and SP1 in the human monocyte leukemia THP-1 cell line and profiled the genome-wide transcriptional response of individual transcription starting sites using deep sequencing based Cap Analysis of Gene Expression. [ J Biol Chem]
CLINICAL RESEARCH
Phase IIB Trial of Oral Midostaurin (PKC412), the FMS-Like Tyrosine Kinase 3 Receptor (FLT3) and Multi-Targeted Kinase Inhibitor, in Patients With Acute Myeloid Leukemia and High-Risk Myelodysplastic Syndrome With Either Wild-Type or Mutated FLT3
The results suggest that midostaurin has hematologic activity in both patients with FLT3-mutant and wild-type. The degree of clinical activity observed supports additional studies that combine midostaurin and other agents such as chemotherapy especially in FLT3-mutant acute myeloid leukemia. [J Clin Oncol]
Phase I/II Clinical Study of Tosedostat, an Inhibitor of Aminopeptidases, in Patients With Acute Myeloid Leukemia and Myelodysplasia
This Phase I/II study demonstrates that oral once daily dosing with 130 mg tosedostat is well tolerated and has significant antileukemic activity. The favorable risk-benefit profile suggests that further clinical trials are warranted. [J Clin Oncol]
LNK Mutation Studies in Blast-Phase Myeloproliferative Neoplasms, and in Chronic-Phase Disease with TET2, IDH, JAK2 or MPL Mutations
LNK mutation analysis was performed in 61 patients with blast-phase myeloproliferative neoplasms; post-primary myelofibrosis in 41, post-polycythemia vera in 11 and post-essential thrombocythemia in 9 patients. [Leukemia]
