SCIENCE NEWS

Preventing Breast Cancer: Largest-Ever Review Completed
The largest review of research into lifestyle and breast cancer ever conducted has confirmed that women can reduce their risk by maintaining a healthy weight, being physically active, drinking less alcohol and breastfeeding their children. [American Institute for Cancer Research and World Cancer Research Fund]

NIH Study Reveals New Genetic Culprit in Deadly Skin Cancer
Researchers detected mutations in the ERBB4 gene (also known as HER4) in 19 percent of patients' tumors, making it by far the most frequently mutated protein tyrosine kinase (PTK) gene in melanoma. This discovery is particularly encouraging because ERBB4 is already targeted by a drug approved for certain types of breast cancer. [National Institutes of Health]

Adjuvant Therapy Benefits Breast Cancer Patients with Small Areas of Cancer in Lymph Nodes
Breast cancer patients with very small areas of cancer in the axillary lymph nodes (micrometastases or isolated tumor cells) appear to benefit from post-surgery treatment with chemotherapy, hormonal therapy, or both. [Susan G. Komen for the Cure]

LABORATORY RESEARCH

The Wnt Receptor, Lrp5, is Expressed by Mouse Mammary Stem Cells and is Required to Maintain the Basal Lineage
This is the first single biomarker that has been demonstrated to be functionally involved in stem cell maintenance. Together, these results demonstrate that Wnt signaling through Lrp5 is an important component of normal mammary stem cell function. [PLoS ONE]

Analyses of Resected Human Brain Metastases of Breast Cancer Reveal the Association between Up-Regulation of Hexokinase 2 and Poor Prognosis
Data suggest that HK2 overexpression is associated with metastasis to the brain in breast cancer and it may be a therapeutic target. [Mol Cancer Res]

ErbB4 Splice Variants Cyt1 and Cyt2 Differ by 16 Amino Acids and Exert Opposing Effects on the Mammary Epithelium In Vivo
Results demonstrate that Cyt1 and Cyt2 ErbB4 isoforms, differing by only 16 amino acids, exhibit markedly opposing effects on mammary epithelium growth and differentiation. [Mol Cell Biol]

Analysis of the Tyrosine Kinome in Melanoma Reveals Recurrent Mutations in ERBB4
These studies could lead to personalized therapeutics specifically targeting the kinases that are mutationally altered in individual melanomas. [Nat Genet]

MicroRNAs and Parallel Stem Cell Lives
Certain microRNAs are downregulated in purified populations of normal mammary epithelial stem cells and breast tumor-initiating cells. These findings have important implications for the regulation of self-renewal and differentiation by microRNAs and suggest new ways of targeting cancer stem cells. [Cell]

CLINICAL RESEARCH

Impact of Micrometastases in the Sentinel Node of Patients with Invasive Breast Cancer
Patients with micrometastatic tumor deposits do not seem to have a worse 8-year disease-free survival or overall survival compared with sentinel node-negative patients. [J Clin Oncol]
