PIK3CA Mutations Associated with Gene Signature of Low mTORC1 Signaling and Better Outcomes in Estrogen Receptor-Positive Breast Cancer
By analyzing gene expression and protein data from nearly 1,800 human BCs, researchers report that a PIK3CA mutation-associated gene signature (PIK3CA-GS) derived from exon 20 (kinase domain) mutations was able to predict PIK3CA mutation status in two independent datasets, strongly suggesting a characteristic set of gene expression-induced changes. CD44pos CD49fhi CD133/2hi Defines Xenograft-Initiating Cells in Estrogen Receptor-Negative Breast Cancer
Researchers show here that both CD44pos CD24neg and CD44pos CD24pos cell populations in estrogen receptor alpha-negative breast tumors are tumorigenic in murine xenograft models. They also describe a third population of xenograft-initiating cells enriched in CD44pos CD49fhi CD133/2hi cells that display heightened tumorigenicity, self-renewal in vivo, and the capacity to give rise to functional and molecular heterogeneity. Phosphodiesterase Type 5 Inhibitors Increase Herceptin Transport and Treatment Efficacy in Mouse Metastatic Brain Tumor Models
These findings suggest that phosphodiesterase 5 inhibitors may effectively modulate blood-brain tumor barrier permeability, and enhance delivery and therapeutic efficacy of monoclonal antibodies in hard-to-treat brain metastases from different primary tumors that had metastasized to the brain.
